Details: A memapsin-2 (β-secretase) inhibitor consisting of the transition-state inhibitor of memapsin-2, OM00-3, (Ki = 0.31 nM) covalently linked to a nine-residue poly D-arginine carrier protein. [OM00-3]r₉ (ELDLpsi[CHOH-CH₂]AVEFGGr₉) was shown to penetrate the plasma membrane of cultured cells and to inhibit production of amyloid β-protein. In transgenic Tg2576 mice with amyloid plaque pathology intraperitoneal injection of N-1920 resulted in a significant decrease in amyloid β-protein levels in plasma and brain.
